Research Interests
The Qin laboratory has been primarily focused on the major projects as follows:
1. One of Dr. Qin’s long-time research focuses is to investigate the roles of the extracellular matrix (ECM), and the non-collagenous proteins (NCPs) in the formation and mineralization of bone, dentin and cementum. Dr. Qin has been focusing on one category of NCPs known as the SIBLING (Small Integrin-Binding LIgand, N-linked Glycoprotein) family, which includes dentin matrix protein 1 (DMP1), dentin sialophosphoprotein (DSPP), bone sialoprotein (BSP), osteopontin (OPN) and matrix extracellular phosphoglycoprotein (MEPE). The Qin laboratory is seeking to understand the overall processes of osteogenesis, dentinogenesis and cementogenesis by focusing upon the nature, metabolism, tissue/cell localization and functions of DMP1, DSPP, BSP and OPN. In particular, DMP1 and DSPP are proteolytically processed into the NH2- and COOH-terminal fragments; Dr. Qin’s group has tested and confirmed the hypothesis that the posttranslational processing events of DMP1 and DSPP play crucial roles in the mechanisms governing the formation and mineralization of bone, dentin and cementum.
2. The second focus of Dr. Qin’s research work is to study the biological roles of FAM20 (Family with sequence similarity 20) members, which include FAM20A, FAM20B and FAM20C. FAM20C is a kinase that phosphorylates many non-collagenous proteins in the mineralized tissues. Mutations in the human FAM20C gene are associated with the hereditary disease known as Raine syndrome. Dr. Qin’s studies on FAM20C have shown that this kinase plays critical roles in biomineralization and regulation of total body phosphorus homeostasis.
3. The third focus of Dr. Qin’s group is to study the nature, tissue/cell localization and functions of bone morphogenetic protein 1 (BMP1) and tolloid-like 1 (TLL1), two proteases that play important roles in the cleaving/activating secretory proteins. Dr. Qin’s group discovered the deletion of Bmp1 and Tll1 in the type I collagen-expressing cells lead to dentin and periodontal defects in mice.
